ABSTRACT

ObjectiveTo investigate the protective effect and mechanism of Achyranthis Bidentatae Radix-Paeoniae Radix Alba on dopaminergic neurons in Parkinson's disease mouse model with the syndrome of ascendant hyperactivity of liver Yang. MethodThe C57BL/6 mice were randomly assigned into normal group, a model group, low-, medium, and high-dose (3.25, 6.5, 13 g·kg-1) Achyranthis Bidentatae Radix-Paeoniae Radix Alba groups, and a selegiline group (0.01 g·kg-1). The mouse model of Parkinson's disease with the syndrome of ascendant hyperactivity of liver yang was established by intragastric administration of Fuzitang combined with intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). The behavioral changes were evaluated by rotarod test and pole test. The protein levels of Ras homolog gene family member A (RhoA), Rho-associated coiled-coil containing protein kinase 2 (ROCK2), myosin light chain 1 (MLC1), and α-synuclein in the substantia nigra were determined by Western blot. Real-time fluorescence quantitative PCR (Real-time PCR) was employed to determine the mRNA levels of RhoA, ROCK2, and MLC1 in the substantia nigra. Enzyme-linked immunosorbent assay (ELISA) was used to measure the levels of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β). The ultrastructural changes of mouse neurons were observed under a transmission electron microscope. ResultCompared with the normal group, the modeling shortened the latency to fall, increased the average total time in the pole test (P<0.01), and up-regulated the levels of RhoA, ROCK2, MLC1, TNF-α, α-synuclein, and IL-1β in the substantia nigra (P<0.05). Compared with the model group, different doses of Achyranthis Bidentatae Radix-Paeoniae Radix Alba and selegiline prolonged the latency to fall, shortened the average total time in the pole test (P<0.05, P<0.01), and down-regulated the levels of ROCK2, MLC1, α-synuclein, TNF-α, and IL-1β in a dose-dependent manner (P<0.05). Further, the modeling decreased the number of cytoplasmic organelles and caused mitochondrial swelling and abnormal shape of endoplasmic reticulum compared with the normal group. The neurons in high-dose Achyranthis Bidentatae Radix-Paeoniae Radix Alba and selegiline groups showed intact nuclei, clear cell boundary, and normal endoplasmic reticulum shape. ConclusionThe combination of Achyranthis Bidentatae Radix and Paeoniae Radix Alba may improve the motor coordination ability of Parkinson's disease mouse model with the syndrome of ascendant hyperactivity of liver yang by inhibiting the neuroinflammation mediated by the RhoA/ROCK2 signaling pathway in the brain.

ABSTRACT

OBJECTIVE To explore the effects of Achyranthis b identatae-Cynanchum otophyllum as core couplet medicinals of Zhengan xifeng decoction on oxidative stress of cerebral tissue in Parkinson ’s disease (PD)mice with syndrome of hyperactivity of liver-Yang. METHODS C57BL/6 mice were randomly divided into normal control group ,model group ,Zhengan xifeng decoction group,A. bidntatae group,C. otophyllum group and couplet medicinals of A. bidentatae-C. otophyllum group,with 10 mice in each group. PD model of hyperactivity of liver -Yang was established by intragastric administration of Aconitum carmichaelii decoction(4 g/kg) and intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (25 mg/kg). The behavioral changes and ultrastructure of substantia nigra neurons in mice were observed . The expressions of tyrosine hydroxylase (TH)positive neurons in substantia nigra were detected . The expressions of total antioxidant capacity (T-AOC),superoxide dismutase (SOD) and malondialdehyde(MDA)in substantia nigra as well as mRNA and protein expressions of nuclear factor -erythroid 2-related factor 2 (Nrf2),thioredoxin reductase 1(Trxr1),thioredoxin interacting protein (Txnip)were also determined . RESULTS Compared with model group ,PD behavior and ultrastructure of substantia nigra neurons were all improved in administration groups . The expressions of TH positive neurons ,T-AOC(except for C.otophyllum group)and SOD (except for C.otophyllum group),mRNA relative expression and protein expression levels of Nrf 2 and Trxr 1 were all increased significantly ;the expression of MDA (except for C.otophyllum group)and mRNA relative expression and protein expression levels of Txnip were all decreased significantly (P<0.05). Intervention effect of couplet medicinals of A. bidentatae-C. otophyllum group was better than that of A.bidntatae group and C. otophyllum group(P< 0.05),and the effect was similar to that of decoction group (P>0.05). CONCLUSIONS The couplet medicinals of A. bidentatae-C. otophyllum can inhibit the level of oxidative stress in the cerebral tissue of PD mice with hyperactivity of liver -Yang by targeting Nrf 2,and play a protective role on the brain neurons . Its effect is better than that of A.bidntatae and C.otophyllum,and it plays the same role as that of the formula in tonifying the kidney ,softening the liver and suppressing the Yang .

ABSTRACT

OBJECTIVE To expl ore the mechanism of effe ctive fractions from Xiongma decoction in the treatment of migraine with hyperactivity of liver-yang and blood stasis. METHODS Totally 70 male SD rats were randomly divided into normal group , model group ,positive control group (Flunarizine hydrochloride capsules 0.9 mg/kg),low-dose and high-dose groups of Xiongma decoction effective fractions (ethyl acetate extract 0.87,3.46 g/kg,n-butanol extract 1.80,7.20 g/kg). Except for normal group , rats in other groups were given aconite decoction (2 g/kg),once a day ,for 4 consecutive weeks to establish the hyperactivity model of liver-yang. On the 15th day of modeling ,all administration groups were given corresponding drugs intragastrically at the same time ,once a day ,for 2 consecutive weeks. On the 29th day of modeling ,rats trigeminal ganglion was stimulated to establish the migraine model with hyperactivity of liver-yang and blood stasis ,then the medication was maintained for another time according to the above method. The macroscopic signs and behavior of the rats were observed ;positive expression ,mRNA and protein expression of transient receptor potential vanilloid 1 (TRPV1),calcitonin generelated peptide (CGRP),calcitonin receptor-like receptor (CRLR) and receptor-associated membrane proteins 1 (RAMP1) in trigeminal cervical spinal complex (TCC) were detected by immunohistochemistry ,RT-qPCR and Western blot assay. RESULTS Rats in model group showed macrophysical signs and behaviora l manifestations related to migraine with hyperactivity of liver-yang and blood stasis. Thirty minutes after last administration ,the above conditions of rats in Xiongma decoction effective fraction groups were improved significantly. Compared with normal group , positive expression,mRNA and protein expression of TRPV 1,CGRP, CRLR and RAMP 1 in TCC of rats in the model group were significantly increased (P<0.05). Compared with model zengguirong@hnse.org group, most of above indicators in Xiongma decoction effective fraction groups were significantly decreased (P<0.05). CONCLUSIONS The mechanism of Xiongma decoction in preventing and treating migraine with hyperactivity of liver-yang and blood stasis may be related to inhibit the activity of TRPV1-CGRP/CGRP receptor signaling pathway in TCC.

ABSTRACT

Objective: To explore the difference of intestinal flora between the patients with acute cerebral hemorrhage due to hyperactivity of liver-Yang and the healthy population. Method: The fecal samples of 9 patients with acute cerebral hemorrhage due to hyperactivity of liver-Yang from the first affiliated hospital of Guangzhou university of traditional Chinese medicine in 2018 were selected as observation group,and 6 stool samples from healthy subjects were selected as the control group.The total bacterial DNA was extracted from the two groups of samples,amplified according to the 16S rRNA V4 region,and paired-end sequencing was performed on the Illumina MiSeq platform.The sequencing results were analyzed by bioinformatics analysis software.The flora composition and structure of the samples from two groups were compared. Result:Venn analysis of operational taxonomic units(OTU) showed significant difference in OTU numbers between the observation group and control group.Partial least squares-discriminant analysis(PLS-DA) showed that there was a significant difference in the composition of intestinal flora between patients with acute cerebral hemorrhage and healthy subjects.On the analysis of species and abundance,at the classification level of phylum,compared with the control group,the ratio of relative abundance values of Firmicutes and Bacteroidetes(F/B) in the observation group was significantly increased,and the relative abundance of Verrucomicrobia was significantly decreased(PPrevotella,Bacteroides,Akkermansia,Blautia and Acidaminococcus(PPBacteroides and Prevotella(B/P) in the observation group was significantly higher than that of the control group;at the classification level of species,there were significant differences between the two groups in P. copri,A. muciniphila,B. ovatus,B. fragilis and Ruminococcus callidus(PPConclusion:Acute cerebral hemorrhage due to hyperactivity of liver-Yang is associated with structural disorder of intestinal flora,which is closely related to the decrease in relative abundance of P. copri and A. muciniphila.

ABSTRACT

Objective To observe the therapeutic effect of DENG Tie-tao's Foot-bath Recipe (mainly composed of Radix Achyranthis Bidentatae,Rhizoma Chuanxiong,Spica Prunellae,Fructus Evodiae,Ramulus Uncariae Cum Uncis,etc.)for the treatment of essential hypertension with hyperactivity of liver Yang syndrome. Methods Eighty essential hypertension patients with hyperactivity of liver Yang syndrome were randomly divided into treatment group and control group,40 cases in each group. Both groups were given basic antihypertensive western medicine treatment, and additionally the treatment group was given external application of DENG Tie-Tao's Foot-bath Recipe and the control group was given foot-bath with warm water. Seven days constituted one treatment course. Before and after treatment , the scores of traditional Chinese medicine(TCM)symptoms and Athens Insomnia Scale were observed , blood pressure variability (BPV)was detected with ambulatory blood pressure monitoring instrument , and plasma norepinephrine(NE) and epinephrine(E) levels were examined by enzyme-linked immunosorbent assay (ELISA). Results (1)The total effective rate for improving TCM symptom scores of the treatment group was 87.5%, and that of the control group was 75.0% , the difference being significant (P < 0.05). (2)The total effective rate for lowering blood pressure of the treatment group was 85.0%, and that of the control group was 72.5% , the difference being significant (P < 0.05). (3)After treatment,scores of Athens Insomnia Scale in the two groups were obviously decreased(P<0.01 compared with those before treatment),and the decrease in the treatment group was superior to that in the control group (P <0.01). (4)After treatment,24h BPV,daytime BPV and night BPV in the two groups were decreased to certain degrees (P < 0.05 compared with those before treatment),and the decrease of 24h BPV and night BPV in the treatment group was superior to that in the control group(P<0.05).(5)After treatment,plasma NE and E levels in the two groups were decreased to certain degree (P < 0 . 05 compared with those before treatment), and the decrease in the treatment group was superior to that in the control group (P < 0.05). Conclusion DENG Tie-Tao's Foot-bath Recipe exerts synergistic action on lowering blood pressure steadily by relieving TCM symptoms, improving the quality of sleep, decreasing 24hBPV of essential hypertension with hyperactivity of liver yang syndrome. And the regulation of plasma NE and E levels may be one of its therapeutic mechanisms.

ABSTRACT

Objective]To summarize Professor Chen Huade’s clinical experience on treatment of hyperactivity of liver yang type vertigo by acupuncture. [Method]By combining with expositions of past dynasties, collecting and analysis of the clinical cases of liver yang type vertigo, to summarize the etiology and pathogenesis of liver yang type vertigo and Professor Chen’s treatment characteristics, and take a case as evidence.[Result]In Professor Chen’s opinion, the basic pathogenesis of hyperactivity of liver yang type vertigo is hepatorenal Yin deficient liver Yang syndrone. Belong to excess in the upper and deficiency in the lower, asthenia in origin and asthenia in superficiality. Based on the theory of “treating disease from the root”,cure this disease by the body acupuncture plus eartip bloodletting therapy, each had a good curative effect.The evidence showed, after the body acupuncture plus eartip bloodletting therapy ten times, vertigo symptom relieved obviously, blood pressure dropped to normal range. In the following up of 3 months, none had a recurrence. [Conclusion]Because of its cheap, non-toxic side effects, significant efficacy, and the characteristics of both symptoms and root causes curing, it is gradually accepted by more and more patients.Professor Chen Huade’s treatment on hyperactivity of liver yang type vertigo by acupuncture has significant effect in clinic which is worth spreading.

ABSTRACT

Objective] To analyse the effect of combining acupuncture of soothing liver, smooth liver and nourishing kidney with Amlodipine Besylate Tablets on light, medium hyperactivity of liver-YANG syndrome hypertension. [Methods] From January 2013 to January 2014, we collected 60 patients with hyperactivity of liver-YANG syndrome meeting the criterion. 30 patients in control group, taking Amlodipine Besylate Tablets, 14 male, 16 female, average age was 62.0 years old(54~72 years old); 30 cases in treatment group, on the basis of control group using soothing liver, smooth liver and nourishing kidney acupuncture, 13 males, 17 females, average age was 61.5 years old(53~73 years old). Patients were collected 24 hours ambulatory blood pressure, anxiety, TCM syndrome integral before and after treatment. [Results] After treatment, control group and the treatment group of ambulatory blood pressure, anxiety and TCM syndrome integral was significantly improved, the differences were statistically significant( P<0.05). In treatment group, daytime systolic blood pressure, daytime diastolic blood pressure, nighttime systolic blood pressure, nighttime diastolic blood pressure, 24h diastolic blood pressure and 24h systolic blood pressure were decreased significantly lower than the control group, and anxiety value, TCM syndrome integral was also improved better than control group(P<0.05). [Conclusion] Soothing liver, smooth liver and nourishing kidney acupuncture combined with oral drug can effectively control blood pressure for hyperactivity of liver-YANG syndrome hypertension patients; meantime it also can significantly relieve the TCM syndrome and anxiety symptom.

ABSTRACT

Objective In the present study,we use GC/MS-based metabolomics approaches to make analysis of serum metabolic profiles of healthy people and the hyperactivity of liver yang type of constitution in patients with essential hypertension.Try to establish the discriminant model,to discover biomarkers (group) of the hyperactivity of liver yang type of essential hypertension,and to explore the essential material basis of Traditional Chinese medicine syndrome theory.Methods Classified according to the guiding principles of Chinese medicine research,the hyperactivity of liver yang type of constitution in male patients with essential hypertension (n=18),as well as health volunteers (n=15) were randomly selected from Guang An Men Hospital clinic,wards and medical center in the first half of 2010,selected patients with essential hypertension requirements are not taking any drugs or Chinese herbs,or stop taking the various drugs more than one week.Extracted Venous Blood of subjects fasting for 12 hours,and serum was separated through centrifugation.Serum samples are stored and at-86℃ refrigerator.Survey and evaluate endogenous metabolism in serum samples of health control group and types of syndrome mentioned above by gas chromatograph mass spectrometry (GCMS)analysis.Then,analyze the metabolites with Partial Least Squares-Discriminant Analysis.Further use PCA to analyze the principal component factor loadings matrix analysis,and for variable scatter plot (Loading plot),significant increase or decrease the variables can be found from the figure.The combination of these variables is the lesion biomarkers group.Results Compared with the health control group,13 differentially expressed metabolites in the essential hypertension hyperactivity of liver yang type group can be identified (P<0.05).8 metabolites were up-regulated expression:Uric acid,citrate,Octadecanoic acid,Hexadecanoic acid,Octadecadienoic acid,Leucine,Cholesterol,Norvaline,and 5 metabolites were down-regulated expression:arachidonate,Oleate,Alanine,Aspartic acid,glycine.Conclusion We are incline to regard that the 13 of EH patient serum differentially expressed metabolites are EH hyperactivity of liver yang syndrome metabolic biomarkers:Uric acid,citrate,Octadecanoic acid,Hexadecanoic acid,Octadecadienoic acid,Leucine,Cholesterol,Norvaline,Arachidonate,Oleate,Alanine,Aspartic acid,glycine.

ABSTRACT

This study was aimed to build metabolic network of hypertension with syndrome of ascendant hyper-activity of liver yang by associating metabolic biomarkers in order to reveal the biological nature of hyperten-sion from the whole view . According to the diagnostic criteria of hypertension with syndrome of ascendant hy-peractivity of liver yang, typical cases were selected and healthy volunteers were enrolled as comparison . Metabolic biomarkers were revealed by metabolomics methods. The metabolic pathways of biomarkers were re-vealed with the KEGG database . The metabolic pathways were connected to establish the metabolic network . Key targets were identified using metabolism node analysis . The results showed that compared with healthy vol-unteers , the metabolic network of hypertension with syndrome of ascendant hyperactivity of liver yang had sig-nificant changes . Norepinephrine , hexanose and arachidonic acid are key network nodes . It was concluded that the metabolic network according to metabolic biomarkers has the advantage in the study on changes of the body under complicated conditions . The study provided a scientific basis for the understanding of biological na-ture of hypertension with syndrome of ascendant hyperactivity of liver yang.

ABSTRACT

Objective To explore the molecular mechanism of the formulae for calming the liver and suppressing YANG in migraine rat model with syndrome of hyperactivity of the liver-YANG.Methods A rat model of migraine with hyperactivity of liver-YANG was established through electrical trigeminal ganglion stimulation and syndrome of oral administration of Fuzi decoction. The total proteins of the lymphocyte in the rats were separated by immobilized pH gradient-based 2-dimensional gel electrophoresis(2-DE), and the 2-DE image was analyzed by PDQuest 7.0 software. Matrix-assisted laser desorption/ionzation-time of flight mass spectrometry (MALDI-TOF-MS) and the SWISS-PORT and MSDB database were used to identify differential proteins.Results The formulae for calming the liver and suppressing YANG could also improve headache. Well-resolution and reproducible 2-DE patterns of rat lymphocyte from normal, model, and therapy tissues were obtained. Eleven of the total 13 differential protein spots were successfully identified by MALDI-TOF-MS. These proteins were alcohol dehydrogenase 3(ADH3), glycogen phosphorylase, ATP synthase D chain, annexin-3, ubiquitin, neutrophil defensin 4 precursor, melanoma-associated antigen E2, heat shock protein-27, annexin-A1, peroxirdoxin-Ⅱ, MU class glutathione S-transferase (Fragment)(GSH). Conclusion Differences occur in the expression of lymphocyte proteins in migraine rats with syndrome of hyperactivity of liver-YANG after treatment with the formulae for calming the liver and suppressing YANG, and the 11 identified protein spots may be associated with its mechanism.

ABSTRACT

Objective To observe the clinical effects of treating Vetigo due to Vertebrobasilar Insufficient blood supply and hyperactivity of liver-Yang with Tianzhi Granules. Methods 118 cases were randomly recruited into a treatment group with 62 cases and a control group with 56 cases. On base of routine treatment, the treatment group was treated with Tianzhi Granules orally; and the control group was treated with Duflomed Hydrochloride orally. One course of treatment was 14 days. Results The total effective rates were 96.77%and 78.57% in treatment group and control group respectively. There was a significant difference between the two groups(P<0.05). The blood flow rat(BFR)of brain was improved in both groups, but the improvement of the BFR in brain was significantly better in the treatment group than the control group (P< 0.05). No adverse reaction was found in the treatment group. Conclusion Tianzhi Granules is effective and safe in treating Vetigo due vertebrobasilar insufficient blood supply.

Objective To explore the molecule mechanisms of migraine with hyperactivity of liver-yang related proteins. Methods Animal model of migraine with hyperactivity of liver-yang was established through electrical trigeminal ganglion stimulation,and by administering orally with Fuzi decoction. The total proteins of splenic lymphocytes in the rats were separated by immobilized pH gradient-based two-dimensional gel electrophoresis. With coomassie blue staining,the 2-DE images were analyzed by PDQuest 7.0 software. Matrix-Assisted Laser Desorption/Ionization-Time of Flight Mass Spectrometry (MALDI-TOF-MS) and database were used to identify the differential proteins. Result Ten differential spots protein were identified,and they were chloride intracellular channel 1,thioredoxin peroxidase Ⅰ,thioredoxin peroxidase Ⅱ,sequence 7 from patent WO9709348,Rho GDP dissociation inhibitor (GDI) alpha,actin related protein 2/3 complex neurofilament light polypeptide,proteasome subunit beta type and heat shock protein-27,annexin A1. Conclusion These protein may be involved in the development of migraine with hyperactivity of liver-yang. These data provide useful clues for elucidating the mechanisms of migraine with hyperactivity of liver-yang.
